Thevasopressin-oxytocinfamilyofpeptidesisofveryancientlineage,foundinorganismsasdiverseashydraandman.Althoughthesepeptideshavebeenintensivelystudiedinvertebrates,thepresumablymoreextensiveinvertebrateserieswasdefinedprimarilybyimmunologicalmethods.Inthisreport,wedescribethepurificationandstructuresoftwopeptidesofthevasopressin-oxytocinfamilyfrommolluscs(“Conopressins”),whichwerefoundinthevenomoffish-huntingmarinesnailsofthegenusConus.TheBIOLOGicalactivityobservedwhenthetwosnailpeptidesareinjectedintracerebrallyintomiceisverysimilartothatelicitedbythevertebrateneurohypophysealhormonesandpresumablyreflectstheiractionsuponacommonreceptorinthebrain.Thesequencesofthepurifiedpeptidesrevealuniquefeaturesnotfoundinthevertebratepeptideseries,mostnotablyanadditionalpositivecharge.Thesearethefirstmembersoftheinvertebrateseriesofthevasopressin-oxytocinfamilytobecharacterizedbiochemically.Thesequencesofthesepeptidesare:fromConusgeographusvenom,Lys-conopressin-G,Cys-Phe-Ile-Arg-Asn-Cys-Pro-Lys-Gly-NH2;andfromConusstriatusvenom,Arg-conopressin-S,Cys-Ile-Ile-Arg-Asn-Cys-Pro-Arg-Gly-NH2.
